Transport across the blood-brain barrier in experimental intracranial meningioma: influence of the degree of tumour angiogenesis and the degree of astrogliosis. Intracranial meningioma is a common intracranial tumour in adults. Recent studies have shown that meningioma angiogenesis, assessed by CD34/CD31 immunohistochemistry, is a strong prognostic factor of local control of the tumour. There are no data on the influence of tumour angiogenesis on other important properties of tumour cells, such as transport across the blood-brain barrier (BBB). In the present study, we performed intravital microscopy to determine the transport of several chemicals across the BBB in experimental meningioma, and we compared the results with those observed in normal brain tissue. Intravital microscopy was performed in the basal brain area of the BBB in the left hemisphere of Fischer-344 rats. In vivo fluorescein transport was performed after giving intraperitoneal injections of FITC-dextran at doses ranging from 20 to 150 microg/g b.wt. Ten minutes later, local concentrations of FITC-dextran in the brain regions distant from the tumour were measured. The pharmacokinetics of mannitol was used to validate transport across the BBB. The concentrations of FITC-dextran in the brain regions distant from the tumour were significantly higher in meningioma than in normal brain tissue at all doses used in the study. In meningioma, the transport of FITC-dextran across the BBB was close to that seen in normal brain tissue, except for the maximal rate of cerebral uptake, which was six times lower in the meningioma. The transport of mannitol across the BBB was significantly higher in meningioma than in normal brain tissue. In addition, the transport of mannitol across the BBB in meningioma was dose-dependently related to the degree of tumour angiogenesis, and it was linearly correlated with the degree of astrogliosis. Parkinson's disease (PD) and essential tremor (ET) have many common symptoms, including parkinsonism and tremor. Quantitative analysis of resting-state slow waves can provide insight into the pathogenesis of PD and other tremor disorders. In the present study, we examined slow-wave characteristics in patients with PD and ET, and the relation between slow-wave parameters and clinical symptoms in the patients with these disorders. We studied 56 consecutive patients with ET, 36 with PD, and 21 healthy control participants. We measured resting-state slow waves, including frequency (delta, theta, alpha), amplitude (power 0.5-1 Hz), and slope (amplitude ratio of 1:50 to 1:25). The amplitude of slow waves in the parkinsonian patients was significantly lower than in the ET patients and control participants. Neither PD nor ET patients showed any significant correlations between slow-wave measures and clinical scores.
